How does magnetic bead-based DNA isolation work?
Magnetic bead-based isolation protocols operate through a series of pipetting steps for lysis, binding, washing and elution – using magnetic separation to isolate the nucleic acids of interest. Unlike traditional spin column-based purification methods, magnetic bead-based workflows do not rely on cumbersome centrifugation or vacuum manifolds to bind and elute nucleic acid, making them ideal for automated DNA isolation protocols.
The Clean Blood & Tissue DNA Kit workflow consists of four easy stages. First, the sample is treated with a lysis buffer to break open cells, release nucleic acids and denature proteins along with other impurities. Next, magnetic beads are added to the lysate, allowing gDNA to selectively bind to the bead surface in the presence of a binding buffer. This is followed by several wash steps designed to remove residual proteins, salts and other contaminants, ensuring high gDNA purity. In the final stage, the purified gDNA is released from the magnetic beads into an elution buffer, making it ready for downstream applications. Heating can be applied during both the lysis and elution phases to enhance DNA yields, offering users added flexibility based on their specific requirements.
Reliable performance across multiple sample types
The Clean Blood & Tissue DNA Kit is engineered to process a wide range of input materials – including blood, buffy coat, tissue and cultured cells – offering high versatility for a range of laboratories. Typical recoveries range from 5 to 7 µg of DNA, and the protocol supports both manual and automated processing of samples. High purity and qPCR-ready DNA
The Clean Blood & Tissue DNA Kit delivers consistent DNA yields with notably high purity:
- OD A260/A230 values are close to the reference range of 2.0 to 2.2, indicating low levels of organic contaminants and salts.
- OD A260/A280 values are consistent with protein-free DNA preparations (~1.8).
- qPCR compatibility was proven by showing no inhibition of signal in the dilution series of the eluate.
